S-Nitrosoglutathione (GSNO), a major intermediate in the nitric oxide (NO)-dependent pathway, is a powerful regulator of energy metabolism. We have investigated the effect of GSNO on ATP, mitochondrial respiration, calcium homeostasis, and cytochrome c release in isolated heart mitochondria subjected to an ischemia-reperfusion (I-R) injury. Isolated hearts from female pigs (31-34 kg) were subjected to 10 min coronary artery ischemia, followed by 10 min reperfusion, and then treated with different concentrations of GSNO (0.1, 1, and 10 microM). In the presence of 1 microM GSNO, ATP content decreased by 10, 24, and 31%, respectively, but was not different in the presence of 0.1 or 10 microM GSNO. GSNO (1 microM) also decreased ADP-stimulated respiration by 26 and 42% in state 4 and 4.8, respectively. GSNO (1 microM) did not decrease the respiratory control ratio or ATP/O ratio. GSNO did not change the state 3 uncoupled respiration. The addition of 10 microM GSNO increased the decrease in state 3 uncoupled respiration caused by ischemia. It was not associated with a change in the time of the occurrence of the ischemia-induced calcium signal or of calcium entry in the mitochondrial matrix. The addition of 1 or 10 microM GSNO did not affect calcium release or calcium uptake in the mitochondrial matrix, but it decreased calcium release from the matrix in the presence of 50 microM carbonylcyanide-m-chlorophenylhydrazone. GSNO also increased the release of cytochrome c to the cytosol in the presence of 50 microM carbonylcyanide-m-chlorophenylhydrazone. The addition of 1 microM GSNO did not have a positive inotropic effect after 10 min of reperfusion.
