The steroidogenic factor-1 (SF-1) gene encodes a protein consisting of 570 amino acids. The major form of the gene product is the SF-1 transcriptional factor; however, an alternate form of the gene encodes a transcriptional repressor. The immediate 5′ region of the mouse gene contains two pairs of perfect consensus hormone response elements that were demonstrated to be functionally active in vitro (Kerrick, et al. (1997) Nature 385:753-760). Other putative elements have also been identified (Chai, et al. (1996) J. Biol. Chem 271:12595-12600). The locus is highly conserved in humans, and the human protein is 89% identical to mouse SF-1.
SF-1 binds to DNA as a monomer and shares a number of functional activities with other members of the nuclear receptor superfamily. For example, SF-1 is a member of the steroid hormone receptor superfamily, which are characterized by the presence of an N-terminal domain of approximately 240 amino acids that is responsible for DNA binding, and a C-terminal domain of varying size that mediates interactions with other transcription factors and other nuclear receptor superfamily members.
SF-1 is expressed at high levels in endocrine tissue, primarily in the testis, but at lower levels in ovary, adrenals, placenta, and hypothalamus (Chakrabarti, (1995) Mol. Reprod
